The Dumas method for protein analysis is a recognized and widely used method for all types of food, feed, and biological materials with low to high protein content. This process begins with the combustion of the sample at a high temperature (approximately 900-1,000 °C) using oxygen gas. Nitrogen in the sample is converted to N₂, separated from other gases with a CO2 carrier, and then measured using a Thermal Conductivity Detector (TCD). Nitrogen content is converted to protein content using a specific conversion factor.
General Requirements:
Protein Analysis using Dumas Method
1. Customers must complete the test form, which can be downloaded from the [Service Files > Service SOP Files] section. The completed form (in .pdf format) must be uploaded along with sample photos in the [Other Supporting Files] and [Photo Data File] sections.
2. Incomplete supporting documents may result in the test sample registration being cancelled by the verifier.
Special Requirements for the KST Gunung Kidul Food Laboratory
1. Analysis must be conducted after collecting a minimum of 20 samples.
2. The method used is based on the total nitrogen content in the sample.
3. Samples must be in solid/powder form with a particle size of ≤ 0.075 mm or ≥ 200 mash.
4. Sample moisture content must be ≤ 15%.
5. Samples must be completely homogeneous, as only a maximum of 250 mg will be sampled (depending on protein content).
6. Provide the protein conversion factor of the candidate sample to be tested and the predicted range of N or protein content in the sample.
