Test principle: DPPH (2,2-diphenyl-1-picryl-hydrazyl-hydrate) is a radical which when dissolved in methanol solvent will have a violet color with an absorption at a wavelength (λ) of 515 nm. Antioxidant testing is based on electron transfer from the analyte (sample containing antioxidants) to DPPH which causes a decrease in the intensity of the violet color of DPPH. The more active the sample, the fainter the violet color will be. The antioxidant activity of the sample was calculated based on the difference in absorbance of the blank solution (only DPPH) and the sample solution + DPPH.
The use of the DPPH assay provides an easy and fast way to evaluate antioxidants by spectrophotometry and can therefore be useful for assessing multiple products at one time."
