Determination of total phenolic compound content in extracts (Total Phenolic Content-TPC). Method: Folin-Ciocalteau (FC) colorimetry is based on the chemical reduction of reagents, a mixture of tungsten and molybdenum oxide based on the method of Singleton, V.L., Orthofer, R., and Lamuela- Raventos, R.M. 1999. Analysis of total phenols and other oxidation substrates and antioxidants by means of Folin-Ciocalteau reagent.

Working principle: The total phenolic compound content in the extract is expressed as gallic acid equivalent (mg/g extract) based on the regression equation of the gallic acid calibration curve. The sample solution (250 μl), standard gallic acid solution (0, 10, 50, 100, and 200 μL) were pipetted into a test tube, then added to 4 mL by adding distilled water and 250 μl Folin-Ciocalteau, the mixture was then shaken until homogeneous. After remaining for 8 minutes, 750 μl of 20% Na2CO3 was added and shaken homogeneously. The mixture was then left for 2 hours at room temperature. Absorption was measured at a wavelength of 765 nm. The measurements were carried out 3 times in repetition so that the phenol content obtained was obtained as equivalent gallic acid (μg/mL sample). The intensity of the blue color formed is proportional to the total phenol content in the sample.
