Ø Colorimetric-AlCl3 Method: Chang, C.C., Yang, M.H., Wen, H.M., and Chernn J.C. Estimation of Total Flavonoids Content in Propolis by Two Complementary Colorimetric Methods, Journal of Food and Drug Analysis. 178-182. 2002.
Ø Prinsif Work: For the determination of total flavonoids, quercetin is used to make a standard calibration curve. Serial standard solutions and samples, each added 2 ml of distilled water. To the tube is added 150 μl NaNO2 5%. After 5 minutes, 150 μl 10% AlCl3 was added. Six minutes later, 2 ml of 1M NaOH and distilled water were added to the volume to 5 ml. The solution was homogeneous and the absorbance measured λ 510 nm with a UV-Vis spectrophotometer. Then the calibration curve is made by connecting the absorption value as coordinates (Y) and the concentration of standard solution as abscissa (X). The obtained flavonoid levels are quercetin equivalent (mg / g extract).
