• Determination of total phenolic compounds in the extract (Total Phenolic Content-TPC)
Ø Method: Folin-Ciocalteau (FC) colorimetry is based on chemical reduction of reagents,
mixture of tungsten and molybdenum oxide based on the method of Singleton, V.L., Orthofer, R., and Lamuela-Raventos, R.M. 1999. Analysis of total phenols and other oxidation substrates and antioxidants by means of Folin-Ciocalteau reagent. Enzymol Methods. 299: 152-178.
Ø Prinsif action: The total phenolic compound content in the extract is expressed as equivalent gallic acid (mg / g extract) based on the regression equation of the gallic acid calibration curve. Sample solution (250 μl), gallic acid standard solution (0, 10, 50, 100, and 200 μL) pipetted into a test tube, then stirred to 4 mL by adding distilled water and 250 μl Folin-Ciocalteau, the mixture then shaken homogeneously. After settling for 8 minutes, 750 μl 20% Na2CO3 is added and shaken homogeneously. The mixture is then allowed to stand for 2 hours at room temperature. Uptake is measured at a wavelength of 765 nm. Measurements were made 3 times so that the phenol content obtained was obtained as equivalent gallic acid (μg / mL sample). The intensity of the blue color formed is proportional to the total phenol content in the sample.
