Research Topic: Application of nanoparticle antibodies to improve the performance of enzyme-linked immunosorbent assay (ELISA) for aplatoxin detection.
Justification:
Aflatoxin is a toxin that is often found in food/feed, especially from agricultural products infected with Aspergillus sp. fungi (A. flavus and A. parasiticus). Aflatoxin contamination is difficult to avoid because tropical climate conditions in Indonesia with high temperature and humidity favour the growth of Aspergillus sp fungi as aflatoxin producers. Aflatoxin is carcinogenic, meutagenic, teratogenic, and immunosuppressive, so its presence in the food chain will endanger human and animal health. Various diseases caused by aflatoxin include liver cancer, weight loss, productivity, and immunity, making the body susceptible to infectious diseases. In addition, the presence of aflatoxins can reduce the quality and acceptability of agricultural products in the global market. Therefore, rapid, sensitive and specific detection techniques need to be developed to support monitoring programmes for aflatoxin contamination in food/feedstuffs, such as ELISA.
Research objectives:
To produce a nanoparticle antibody-based ELISA kit to detect aflatoxin contamination in food/feed, with the following targets:
1. To produce a validated prototype of anti-AFB1 nanoparticle antibody-based ELISA kit,
2. Obtained homogeneity test data from the ELISA kit produced,
3. Obtained ELISA kit stability test data during the storage period,
4. Obtained ELISA kit validation data for detecting aflatoxin in food/feed.
Scope of Activities:
1. Antibody production and purification
2. Testing of nanoparticle antibody-based ELISA system,
3. Assembly of aflatoxin ELISA kit components,
4. Homogeneity test and stability test of ELISA kit,
5. Validation of nanoparticle antibody-based ELISA kit,
6. Detection of aflatoxin contamination in feed and food ingredients.
