This study aims to test the effectiveness of Moringa oleifera leaves and turmeric (Curcuma longa) as natural antimicrobials in controlling mastitis in dairy cows, especially against Staphylococcus aureus bacteria. This test is needed to determine the effective dose of extract in killing and inhibiting the growth of bacteria that cause mastitis. In addition, this study also evaluates the application of biosecurity at the farmer level as part of a holistic disease control approach.
The initial stage of the study began with taking cow's milk samples from the field, which were then tested using the California Mastitis Test (CMT) method. Milk samples that showed positive results were then taken as much as 2 ml for the bacterial isolation process. Isolation was carried out by culturing samples on BHI broth media and incubating for 24 hours at 37°C. Furthermore, subculture was carried out using Baird Parker agar media, and re-incubated for 24 hours. Black colonies with clear zones around them were identified as S. aureus, then confirmed through DNA extraction using a DNA extraction kit.
Moringa leaf extract was obtained using the Nfambi et al. method. (2015) and Zhu et al. (2020), while turmeric extract was obtained using a Soxhlet apparatus according to the method of Naz et al. (2010). Antibacterial activity testing was carried out using the disc diffusion method on BHI agar media, where the biogram disc that had been dripped with moringa leaf extract and turmeric was placed in the center of the agar plate. After incubation, the inhibitory power was assessed based on the formation of a clear zone around the disc. As part of an integrative approach, interviews were also conducted with farmers using questionnaires to assess the level of biosecurity implementation at the livestock location. The activity locations include Bogor City, Bogor Regency, Depok, East Jakarta.
